rProtein A Sepharose™ 4 Fast Flow est un milieu d'affinité aux anticorps bien établis, conçu pour la purification des anticorps monoclonaux et polyclonaux basée sur la plateforme Sepharose™ Fast Flow. La protéine A recombinée est produite dans E.coli et est synthétisée pour un couplage orienté afin de donner une matrice ayant une capacité de liaison accrue. Le couplage basé sur les groupes époxy assure une fuite réduite des ligands. La spécificité de la protéine A recombinée pour la région Fc de l'IgG est similaire à la protéine A originelle et elle donne une excellente purification en une seule étape. It is composed of recombinant protein A coupled to the cross-linked 4% agarose base matrix. The recombinant protein A is produced in E. coli and has been specially engineered to favour an oriented coupling giving a matrix with enhanced binding capacity. The epoxy based coupling ensures low ligand leakage. The specificity of the recombinant protein A for the Fc region of IgG is similar to native protein A, giving a high purification factor in a single step. The high capacity, low ligand leakage and a well established base matrix makes rProtein A Sepharose 4 Fast Flow suitable for purification of monoclonal antibodies at both laboratory and process scale.
