La PCR est une méthode utilisée pour amplifier une séquence d'ADN spécifique qui est généralement amplifiée dans une réaction contenant une ADN polymérase thermostable, des nucléotides et des amorces complémentaires de la séquence cible. Lorsque cette solution est chauffée, la molécule d'ADN se dénature et se sépare en deux brins. À mesure que la solution refroidit, les amorces se fixent aux séquences cibles par annelage dans les brins d'ADN séparés, et la polymérase d'ADN synthétise un nouveau brin en étendant les amorces à l'aide de nucléotides, créant ainsi une copie de la séquence d'ADN (amplicons). Lorsqu'il est répété, ce cycle de dénaturation, de recuisson et d'extension augmente de façon exponentielle le nombre d'amplicons cibles. Dans la PCR en temps réel, les sondes fluorescentes spécifiques sont utilisées pour détecter l'ADN amplifié par hybridation avec les amplicons. Ces sondes sont liées à un fluorophore à une extrémité et à un suppresseur (quencher) qui supprime la fluorescence à une autre. Si la séquence cible est présente pendant la PCR, une amplification se produit et la sonde est dégradée, provoquant ainsi une augmentation de la fluorescence. La fluorescence est mesurée par un détecteur et le logiciel associé trace l'intensité de la fluorescence par rapport au nombre de cycles, permettant ainsi de déterminer la présence ou l'absence de l'organisme cible.

Conformément à la Directive de la commission européenne 2002/86/CE, les ingrédients alimentaires doivent être déclarés. Dans le cas de viande et de produits à base de viande, les espèces animales doivent être mentionnées sur l'étiquette. En outre, l'authenticité des espèces peut être extrêmement importante pour les consommateurs pour des raisons économiques, médicales, culturelles et religieuses. L'utilisation frauduleuse de viandes bon marché à la place d'espèces plus onéreuses, l'ajout de viande dans des produits (végétariens) non carnés et la présence d'allergènes dans les denrées alimentaires témoignent clairement de l'importance du problème (Ballin, 2010 ; Dooley et al., 2004).
Plusieurs méthodes sont basées sur l'identification de protéines au moyen de procédés électrophorétiques et/ou immunologiques. Cependant, ces méthodes n'offrent aucune fiabilité lorsqu'elles sont appliquées à des produits hautement transformés et chauffés en raison de la détérioration des protéines. L'ADN est plus stable que les protéines pendant le traitement et même s'il peut être fragmenté en plusieurs procédés, des méthodologies génétiques modernes telles que les techniques basées sur la PCR permettent toujours l'identification de l'ADN à partir de différentes espèces présentes dans un échantillon (Lockley & Bardsley, 2000). Les techniques de PCR en temps réel sont particulièrement adaptées à ces produits, car les petits fragments d'ADN peuvent toujours être amplifiés et identifiés avec une sensibilité et une spécificité élevées (Dooley et al., 2004).
