PCR barcoding expansion

PCR Barcoding Expansion packs enable multiple nanopore sequencing libraries to be pooled and run on a single MinION Flow Cell.

PCR Barcoding Expansion packs are designed to allow the pooling and running together of multiple sequencing libraries on nanopore sequencing devices for reduced per sample sequencing costs.

The kits include adapters and Oxford Nanopore forward and reverse PCR primers with the same barcode in both forward and reverse primers. The PCR Barcoding Expansion 1-12 offers 12 unique barcodes and enough of the required adapters to make 6 libraries per barcode, while the PCR Barcoding Expansion 1-96 offers 96 unique barcodes and enough of the required adapters to make 10 libraries per barcode. The barcodes have been carefully designed and extensively purified to minimise the opportunity for cross-talk.

Deconvolution of barcoded sequencing data is supported by the MinKNOW software provided with nanopore sequencing devices, which classify the barcode sequence and sort reads into corresponding folders.

These kits should be used with the Ligation Sequencing Kit for library preparation.

Note: There are currently two protocols provided with the kits as examples of how the PCR Barcoding Expansion packs could be applied for genomic and amplicon libraries. If you are barcoding cDNA or amplicons, these have to be pre-made in advance. These protocols are open for adaptation and optimisation for specific experimental requirements.

Sample multiplexing for PCR Barcoding Expansion 1-12 is ≤12 and PCR Barcoding Expansion 1-96 is ≤96.

Lieferinformationen: Flow cells and kits are shipped together at 2 to 8°C. Upon receipt, please place the product in a long-term storage location at –20°C. Oxford Nanopore Technologies deem the useful life of the product to be 3 months from receipt by the customer.
