VWR® Trypsin-EDTA solutions are used in cell culture to remove adherent cells from a culture surface. Trypsin is a porcine pancreas-derived enzyme that is commonly used for the dissociation and disaggregation of anchorage-dependent mammalian cells and tissues. The concentration of trypsin necessary to separate cells from their substrate is dependent primarily on the cell type and the age of the culture. EDTA, a chelating agent, enhances enzymatic activity by removing calcium and magnesium ions. These ions obscure the peptide bonds on which trypsin acts as well as enhancing cell to cell adhesion.

Phenol red is used as a pH indicator in cell culture. Its colour exhibits a gradual transition from yellow to red over the pH range 6,8 to 8,2. Above pH 8,2, phenol red turns a bright pink (fuchsia) colour.

Nach dem Auftauen kann das Passagekit bei 2 °C bis 8 °C im Dunkeln bis zu 1 Monat gelagert werden.. Das Medium nicht über 37 °C erhitzen oder unkontrollierbare Wärmequellen (z. B. Mikrowellengeräte) verwenden.
Frieren Sie das Kit nicht wieder ein. Dies kann zu hohen Salzkonzentrationen durch Ausfrieren von reinem Wasser führen, was zu irreversiblen Schäden führt.
Übermäßige Trypsinierung führt zu irreversiblen Schäden.
Überschreiten Sie aufgrund der tryptischen Aktivität dieses Passagekits nicht die empfohlene Inkubationszeit. Wenn die Zellen zu lange der Inkubation ausgesetzt werden, führt dies zu irreversiblen Schäden an Ihrer Kultur.

Das Provitro-Wachstumsmedium für Skelettmuskelzellen ist ein steriles flüssiges Kulturmedium zur Kultivierung menschlicher Skelettmuskelzellen (HSKMC). Das Medium wird als Basalmedium geliefert und ist nach Zugabe der Ergänzungskit-Komponenten zur Kultivierung von Provitro HSKMC geeignet. Die Formulierung ist für eine anfängliche Anzucht von 4000 Zellen/cm² bis zur Konfluenz (ca. 90%) optimiert. Futterschicht, Matrixsubstrate oder andere Substanzen sind nicht erforderlich. Aufgrund der Möglichkeit einer verringerten proliferativen Aktivität empfehlen wir, das Antibiotika-Ergänzungsmittel nur für frisch isolierte Zellen zu verwenden.
