Aufreinigung und Größen-Bestimmung von grün und von blau fluoreszierenden Proteinen

Wenn zur Herstellung medizinisch brauchbarer Proteine durch Transformation Bakterien verwendet werden, müssen die fraglichen Proteine von allen anderen zellulären Proteinen getrennt werden. In diesem Experiment dienen die einmaligen Fluoreszenzeigenschaften des GFP und des BFP als Test im Verlauf der Aufreinigung der aus einem E. coli-Extrakt. Die Säulenfraktionen, die GFP oder BFP enthalten, werden durch Fluoreszenz identifiziert und anschließend aufgereinigt. Im Rahmen einer optionalen Übung können die aufgereinigten Proteinfraktionen durch SDS-Polyacrylamidgel-Elektrophorese (SDS-PAGE) getrennt werden, damit sich Reinheit und Größe der GFP und BFP einschätzen lassen.

The Gram staining method was first described in 1844 by the Danish bacteriologist Hans Christian Gram, after whom the test was named.
