TPO is a lineage-specific growth factor produced in the liver, kidney and skeletal muscle. It stimulates the proliferation and maturation of megakaryocytes, and promotes increased circulating levels of platelets in vivo. TPO signals through the c-mpl receptor, and acts as an important regulator of circulating platelets. Human and murine TPO exhibit cross-species reactivity. The human TPO gene encodes for a 353 amino acid glycoprotein, which contains a 21 amino acid signal sequence, a 15 amino acid erythropoietin-like domain, and a highly glycosylated 179 amino acid C-terminal domain. Recombinant Human TPO is a fully biologically active 174 amino acid polypeptide (18.6 kDa), which contains the erythropoietin-like domain of the full length TPO protein.

RANKL and RANK are members of the TNF superfamily of ligands and receptors that play an important role in the regulation of specific immunity and bone turnover. Defensins (alpha and beta) are cationic peptides with a broad spectrum of antimicrobial activity that comprise an important arm of the innate immune system. The alpha-defensins are distinguished from the beta-defensins by the pairing of their three disulfide bonds. To date, six human beta-defensins have been identified; BD-1, BD-2, BD-3, BD-4, BD-5 and BD-6. beta-defensins are expressed on some leukocytes and at epithelial surfaces. In addition to their direct antimicrobial activities, they can act as chemoattractants towards immature dendritic cells and memory T cells. The beta-defensin proteins are expressed as the C-terminal portion of precursors, and are released by proteolytic cleavage of a signal sequence and, in some cases, a propeptide sequence.
