There are two kinds of IGF receptors based on their relative affinities for IGFI versus IGFII. The IGFI receptor prefers IGFI over IGFII and weakly binds insulin. IGFI receptor is a disulfide-linked heterotetrameric transmembrane protein consisting of two alpha (130 kD) and two beta (95 kD) subunits. Both the alpha and beta subunits are encoded within a single receptor precursor cDNA. The IGFI receptor is therefore similar in structure to the insulin receptor. The proreceptor polypeptide is proteolytically cleaved and disulfide-linked to yield the mature heterotetrameric receptor. The alpha subunit of the IGFI receptor is extracellular while the beta subunit has an extracellular domain, a transmembrane domain and a cytoplasmic tyrosine kinase domain. The IGFI receptor is highly expressed in all cell types and tissues. Essentially all of the biological activities of IGFI and II have been shown to be mediated via the IGFI receptor.

The c-Myc protein is a transcription factor, which is encoded by the c-Myc gene on human chromosome 8q24. c-Myc is commonly activated in a variety of tumor cells and plays an important role in cellular proliferation, differentiation, apoptosis and cell cycle progression. The phosphorylation of c-Myc has been investigated and previous studies have suggested a functional association between phosphorylation at Thr58/Ser62 by glycogen synthase kinase 3, cyclin-dependent kinase, ERK2 and C-Jun N-terminal Kinase (JNK) in cell proliferation and cell cycle regulation. Studies also have shown that c-Myc is essential for tumor cell development in vasculogenesis and angiogenesis that distribute blood throughout the cells, and which brought extensive attention in the development of new therapeutic approach for cancer treatment.

The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. N ethylmaleimide sensitive factor (NSF) is an ~83 kDa protein (predicted MW) which was initially isolated based on its ability to restore intercisternal Golgi transport to N ethylmaleimide treated membranes. NSF was subsequently discovered to be involved in a variety of membrane fusion events. NSF is an ATPase with two ATP binding domains that are essential for membrane fusion and homo oligomerization. In order to interact with target membranes, NSF requires another set of soluble proteins called Soluble NSF attachment proteins (SNAPs). NSF has been shown to play a prominent role in synaptic vesicle fusion. Together with synaptotagmin and a SNAP, NSF modulates the interaction of SNAP25, VAMP, and syntaxin, in an ATP dependent manner, to form a complex with a sedimentation coefficient of 20 S. This complex may represent an intermediate involved in synaptic vesicle docking and fusion.

CNTF is a survival promoting factor for different types of neurons in vitro and in vivo.
