Myogenin is a member of a family of myogenic regulatory genes, which includes MyoD, myf5 and MRF4. These genes encode a set of transcription factors, which are essential for muscle development. Transfection of myogenin into multipotential mesodermal cells have been shown to convert the mesodermal cells to myoblasts. Expression of myogenin is restricted to cells of skeletal muscle origin. It is therefore a useful marker for tumors of the muscle lineage, being strongly expressed in alveolar rhabdomyosarcomas.

The protein encoded by this gene is a component of the exocyst complex, a multiple protein complex essential for targeting exocytic vesicles to specific docking sites on the plasma membrane. Though best characterized in yeast, the component proteins and functions of the exocyst complex have been demonstrated to be highly conserved in higher eukaryotes. At least eight components of the exocyst complex, including this protein, are found to interact with the actin cytoskeletal remodeling and vesicle transport machinery. RAC, a small GTPase in the ras family, regulates the formation of membrane ruffles, lamellipodia and filopodia. It is involved in cytoskeletal actin organization and transformation. The p21 activated kinases (PAKs) are direct targets of active Rac and Cdc42 which can induce the assembly of polarized cytoskeletal structures when expressed in fibroblasts. RAC1 and RAC2 are also essential components of NADPH oxidase, the enzyme responsible for generating free radicals.

Platelet-activating factor (PAF) is a biologically active phospholipid with diverse biologic effects. PAF is degraded to inactive products by hydrolysis of the acetyl group at the sn2 position to produce the biologically inactive products LYSO PAF and acetate. This reaction is catalyzed by PAF acetylhydrolase (PAFAH). The various monomeric and multimeric forms of the enzyme are composed of alpha, beta, and gamma PAFAH subunits. The catalytic activity of the enzyme resides in the beta and gamma subunits, whereas the alpha subunit has regulatory activity. Trimer formation is not essential for the catalytic activity.

PCNA, also known as polymerase delta auxiliary protein, is essential for DNA replication and is involved in DNA excision and mismatch repair pathways. PCNA binds to the CDK inhibitor p21, the structure-specific endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT). PCNA is a potentially useful marker of cells with proliferative potential and for identifying the proliferation status of tumour tissue (i.e. relevant to prognosis).
