The RM134L monoclonal antibody specifically reacts with mouse CD252, a TNF/NGF superfamily member present on activated B lymphocytes and antigen-presenting cells. On activated B cells it enhances immunoglobulin secretion and cell proliferation. CD252 is also known as the OX-40 ligand and interacts with the OX-40 antigen found on the surface of activated T cells. The RML134L is reported to block the costimulatory activity of OX-40L.

The 53-6.7 monoclonal antibody specifically reacts with Ly-2, the 38 kDa alpha chain, and with Lyt-2, the 34 kDa alpha’ chain, of the mouse CD8 antigen. The alpha’ chain is the truncated form of alpha chain, encoded by the same CD8a gene. In CD8a, the alpha and alpha’ chains form heterodimers with CD8b (the beta chains) or homodimers (alpha-alpha), which occur as receptors on the surface of the majority of thymocytes. The UCHT1 monoclonal antibody specifically reacts with the ε chain of the CD3/T lymphocyte antigen receptor complex. The CD3 complex contains γ, δ, and ε chains, and it is part of the TCR complex, expressed by all mature T lymphocytes and by the thymocyte lineage. CD3 enhances the antigen recognition by signal transduction. Unlike HIT3a, another specific antibody of CD3, the UCHT1 antibody can stain both the surface and intracellular CD3ε. The immobilized UCHT1 can cross-link with the TCR complex, enhancing cellular activation and proliferation.

The O323 monoclonal antibody specifically binds to the human CD27, which is a lymphocyte-specific member of the TNF/NGF-R family, expressed on a subset of human thymocytes and the majority of the mature T cells. CD27 is upregulated by T lymphocytes stimulation, and binds to CD70, enhancing the interaction between T and B lymphocytes. CD27 is a marker of memory B cells.

The M5/114.15.2 monoclonal antibody or (M5/114) is specific for the mouse major histocompatibility complex class II, specific for the I-A and I-E subregion-encoded glycoproteins (alloantigens I-Ab, I-Ad, I-Aq, I-Ed, and I-Ek, but not the I-Af, I-Ak, nor I-As antigens). The receptor for the B7-H1 molecule is PD-1, which contains an Immunoreceptor Tyrosine-based Inhibitory Motif (ITIM), and is expressed on activated B and T cells. The RPA-T8 monoclonal antibody specifically reacts with the human CD8a molecule, a 32 kDa cell surface receptor expressed either as a heterodimer (CD8 α/β) or as a homodimer (CD8 α/α) on the majority of thymocytes, a subpopulation of mature T cells, and natural killer cells. CD8 interacts with the major histocompatibility complex class I (MHC class I) molecules on antigen-presenting cells or epithelial cells. The RPA-T8 antibody reacts with 13-48% of peripheral lymphocytes, 80% of thymocytes, and a subset of natural killer cells. RPA-T8, OKT8, and HIT8a antibodies do not compete with each other for binding to peripheral leukocytes, meaning that that they do not recognize the same epitope or block each other by steric hindrance.
