PureCube amine carboxy agarose resin is your best option to couple biomolecules with free-standing amine (NH₂) groups.
- Usage: Specific binding and purification of Rho1D4-tagged proteins
- Specificity: Affinity to Rho1D4-tagged (TETSQVAPA) proteins
- Binding capacity: >3 mg/ml
- Bead Ligand: anti-Rho1D4 antibody
- Bead size: 400 μm
- Matrix: 7.5% cross-linked agarose
- Filling quantity: Delivered as a 50% suspension
- Short-term storage: In neutral buffer at 4 °C
- Long-term storage: In neutral buffer with 20% ethanol at 4 °C
- Notes about FPLC use: low pressure only
Required equipment:
- Lysis buffer
- Wash buffer
- Elution buffer (incl. Rho1D4 peptide)
- Ice bath
- Refrigerated centrifuge for 50 ml tube (min 10000×g)
- 50 ml centrifuge tube
- Micropipettor and micropipetting tips
- Disposable gravity flow columns with capped bottom outlet, 2 ml
- pH meter
- End-over-end shaker
- SDS-PAGE buffers, reagents and equipment optional: Western Blot reagents and equipment
The resulting peptide bond between the carboxy agarose beads and your biomolecule of interest is stable so that the bead can be used for other applications afterward. Our PureCube carboxy activated agarose is also available as magnetic beads if you prefer this purification method or as XL agarose resin if you need higher flow rates for your downstream application.
