phi29-XT RCA Kit
The phi29-XT RCA DNA amplification kit includes everything needed for sensitive and robust rolling circle amplification (RCA), an isothermal amplification approach to continuously amplify circular DNA by generating long, repetitive copies of the circular sequence.
- High sensitivity: amplify from as little as 1 fg of input plasmid DNA
- Powered by phi29-XT DNA Polymerase, an engineered polymerase
- Simple workflow produces high yield in a short reaction time
- Includes exonuclease-resistant random primers and dNTPs

RCA enables several downstream applications, including DNA sequencing, cell-free protein expression, viral DNA enrichment, plasmid amplification, and padlock probe-based assays. The kit features phi29-XT DNA polymerase, an engineered polymerase with improved thermostability, sensitivity, and capable of generating higher yield in a shorter reaction time than wild-type phi29 DNA polymerase.
Also included are exonuclease-resistant random primers (containing phosphorothioate bonds) to universally amplify any circular DNA sequence and dNTPs. Input material can be a purified plasmid, liquid media culture, a cultured colony from an agar plate, or a glycerol stock.
