Proteinase K is a stable serine protease with broad substrate specificity and will degrade many proteins in their native conformation, even in the presence of detergents (e.g., SDS).

- Unit Definition: One unit (U) of enzyme will hydrolyze urea-denatured hemoglobin to produce 1.0 μmole of tyrosine per minute at pH 7.5 at 37°C.
The enzyme is frequently used in molecular biology applications to digest unwanted proteins such as nucleases from DNA and/or RNA preparations from microorganisms, cells, and plants.
