Nuclear factor erythroid 2-related factor 2 (Nrf2) is a transcription factor, which regulates the expression of many detoxification and antioxidant enzymes. Nrf2 can potentially play a significant role in adaptive responses to oxidative stress. Nrf2 belongs to the Cap N Collar subfamily of basic leucine zipper (CNC-bZIP) transcription factors. The NF-E2 related family comprises of Nrf1 (TCFII), Nrf2, and Nrf3 and these proteins heterodimerize with Maf proteins to bind to Maf recognition element (MARE). Under normal conditions, Nrf2 is inactive and localized in the cytoplasm through its association with Keap1. When cells are exposed to electrophiles or oxidative stress generating agents cytoplasm localization of Nrf2 is inactivated and Nrf2 translocates into the nucleus and binds to specific DNA sequence elements known as NF-E2 binding site, Maf recognition element (MARE), stress response element or anti-oxidant response element (ARE). Studies have investigated the role of Nrf2 in urinary bladder carcinogenesis, pulmonary fibrosis, and smoke-induced emphysema. A study reported Nrf2-mediated ARE transcription to be a likely target of preventative therapy in neurodegenerative disorders such as Huntington's disease.

CD138, also known as Syndecan-1, is a member of the transmembrane heparan sulfate proteoglycan family, acts as an extracellular matrix receptor and is involved in many cellular functions, including cell-cell adhesion and cell-matrix adhesion. CD138 expression is found in both hematopoietic and non-hematopoietic cells. In the hematopoietic system, CD138 labels plasma cells. It is an excellent marker for plasmacytic differentiation within the spectrum of hematologic malignancy. Among non-hematolymphoid cells, CD138 reactivity is observed in many types of epithelial cells and stoma cells in both normal and tumor tissues.

This antibody reacts with a 34 kDa protein known as calponin. Calponin, a calmodulin, binds tropomyosin and F-actin and is thought to be involved in the regulation of smooth muscle contraction. Expression of calponin is restricted to smooth muscle cells. Two isoforms of calponin exist whose molecular weights are 34 kDa and 29 kDa. 29 kDa isoform is primarily restricted to the muscle of the urogenital tract.

Calcitonin gene-related peptide (CGRP) is a 37-amino acid disulphide-bridged sensory neuropeptide present in the central and peripheral nervous systems of many animal species. Two forms of CGRP (α and β) have been identified from both rat and human, and the sequence of chicken CGRP has also been reported, showing striking structural similarity to the mammalian forms. CGRP is abundant in perivascular sensory neurons and is known to be a potent vasodilator. Recent work supports the rôle of CGRP in mediating skin vasodilatation and differential mesenteric artery vasorelaxation, as well as serving as an endogenous myocardial protective substance with a possible involvement in the inhibition of human platelet aggregation. CGRP is frequently co-localised with Substance P and immunoelectron microscopic observations have shown that the peptides are co-stored in single secretory granules both in sensory neurons and in thyroid C-cells. There are many hundreds of reports in the scientific literature describing the localisation and co-existence of CGRP with other transmitter molecules in a vast array of mammalian and non-mammalian species.

The adenovirus early gene product E1A is a potent stimulator of cellular proliferation, which when overexpressed can overcome the growth-inhibitory effects of TGF-β. The E1A region encodes a series of related proteins (35 to 46 kDa) with multifunctional capabilities and forms a specific complex with the retinoblastoma tumor suppressor gene product. The E1A and E1B regions together comprise the transforming region of adenovirus. While expression of E1A alone is sufficient to immortalize primary cells, complete transformation requires the additional expression of the E1B region.

This complex provides antigen-specific recognition and subsequent activation of B cells to proliferate and differentiate into antibody-secreting cells (plasma cells) or memory B cells, which are crucial for secondary antigen encounter. The EBVCS-5 antibody is specific for human CD23, also known as the low affinity IgE receptor (FceRII), a 45 kDa type II transmembrane protein and member of the immunoglobulin superfamily. It is expressed on most B cells and is upregulated upon activation. It is also present on mantle zone B cells, eosinophils, monocytes and a subset of T cells and platelets. CD23 plays a role in B cell development and differentiation, and also functions to regulate IgE production. Soluble secreted forms of CD23 have been reported to bind IgE, stimulate release of pro-inflammatory cytokines from monocytes, and play a role in B cell differentiation.

The lymphocyte activation gene-3 (LAG-3, CD223), a member of the immunoglobulin superfamily (IgSF) related to CD4, binds to the major histocompatibility complex (MHC) class II molecules but with higher affinity than CD4. Several alternative mRNA splice-variants of human LAG-3 have been described, two of them encoding potential secreted forms: LAG-3V1 (i.e. the D1-D2 domains of the protein, 36 kDa) and LAG-3V3 (D1-D3, 52 kDa). The longer form was detected by ELISA in the serum of healthy individuals as well as of tuberculosis patients with a favorable outcome. LAG-3 expression by T cell clones correlated with IFN-γ production, and hence soluble LAG-3 has been suggested as a serological marker of Th1 responses.
