Uroplakins (UPs) are a family of transmembrane proteins (UPs Ia, Ib, II and III) that are specific differentiation products of urothelial cells. Uroplakins are markers of terminally differentiated urothelium. Uroplakin II (UPII) is a newly described sensitive marker for urothelial carcinoma (UC). The expression profile of UPII in different types of UC and its utility in the diagnostic setting are needed.

The ACK2 antibody is specific for CD117, also called c-Kit, a 145 kDa cytokine receptor important in the development of hematopoietic stem cells, in oogenesis, and for functional activity of immune cells such as NK and mast cells. c-Kit binds to a ligand known as stem cell factor (SCF), or alternatively as mast cell growth factor. Ligand binding promotes the activation (dimerization) and subsequent tyrosine kinase activity of the c-Kit receptor and triggers key survival, expansion and maturation signals during hematopoietic progenitor cell development. Conversely, shedding of extracellular domain of c-Kit receptor is reported to induce inactivation or apoptosis within these cells. The survival signaling activity of c-Kit confers a proto-oncogenic attribute to the receptor, as overexpression or mutations in this protein are associated with tumor development.

The RM134L antibody recognizes CD252, also known as OX-40 Ligand or CD134 Ligand, a member of the TNF superfamily that is present on the surface of antigen presenting cells and activated B lymphocytes. The OX-40 Ligand interacts with OX-40 (CD134) which is expressed primarily on activated T cells. This costimulatory interaction leads to increased proliferation and IL-2 production responses of activated T cells, and at the same time enhances proliferation and immunoglobulin secretion by activated B cells.

p63 is a homolog of the tumor suppressor p53, it is identified in basal cells in the epithelial layers of a variety of tissues, including epidermis, cervix, urothelium, breast and prostate. p63, vital for the development of the prostate and selectively expressed by normal prostate basal cells, has been shown to be a promising complementary basal cell specific marker to High Molecular Weight-Cytokeratin (HMW-CK), for the differential diagnosis of benign prostatic lesions and prostatic carcinoma. p63 has also been shown to be a sensitive marker for lung squamous cell carcinomas (SqCC), with a sensitivity of ~90% . Specificity for lung SqCC, vs. lung adenocarcinoma (LADC), is approximately 80. In breast tissue, p63 has been identified in myoepithelial cells of normal ducts.
