The M5/114.15.2 antibody reacts with mouse MHC Class II alloantigens I-Ab, I-Ad, I-Aq, I-Ed, and I-Ek, as well as being cross-reactive with mouse cells of H-2p and H-2r haplotype. MHC Class II is widely expressed by mouse immune cells bearing these alloantigens, including T and B cells, monocytes, macrophages and dendritic cells. The antibody does not react with the following alloantigens: I-Af, I-Ak, I-As, or NOD H-2g7.

The MF23 antibody reacts with mouse Foxp3, a 50-55 kDa transcription factor which is a central regulator of T cell activity and is critical for the development and function of regulatory T cells (Tregs). Foxp3 is a member of the forkhead box or winged helix family of transcription factors and is expressed at constitutively high levels in Treg cells. Forced expression of Foxp3 in conventional T cells results in the upregulation of Treg associated molecules such as CD25, CTLA-4, and GITR and is sufficient to impart suppressive functional activity to these cells.

The NOK-1 antibody reacts with human CD178 (Fas ligand) in both membrane bound and soluble forms. Fas ligand is a 40 kDa transmembrane glycoprotein, a member of the TNF family, and is expressed by activated T and NK cells, neutrophils, and monocytes. Interactions between CD178 (Fas ligand) and CD95 (Fas) induce a program of apoptosis and play a key role in immune regulation and homeostasis. The extracellular domain of human CD178 can be cleaved from the surface by matrix metalloproteinases (MMPs) resulting in a 26 kDa soluble protein.

Monoclonal Anti-BCL-10 reacts specifically with human bcl-10. The epitope recognized by the antibody resides within amino acids 122-168 of human bcl-10 molecule. BCL-10, also designated as CIPER, mE10, cE10, CARMEN, and CLAP, is an N-terminal CARD (caspase recruitment domain) containing protein. It is a cellular homologue of the equine herpesvirus-2 protein E-10 (VCLAP). BCL-10 was implicated in the regulation of apoptosis by interacting with caspase 9, enhancing procaspase 9 processing, and triggering its activation when overexpressed in the cell. Bcl-10 cellular overexpression induces JNK, P38, and NF-kB activation. Deregulation of BCL-10 expression was also demonstrated to be involved in cellular oncogenesis.

The BM8.1 antibody is specific for mouse F4/80 antigen, a 125 kDa transmembrane protein widely expressed by members of the mononuclear phagocyte system and considered to be a key marker for mature macrophage cells. F4/80 is differentially expressed during myeloid cell development, and may be regulated by certain cytokines within the tissue microenvironment. Other cell types shown to express this antigen include Langerhans cells, Kupffer cells and dendritic cell subsets. BM8.1 is widely used together with antibodies to CD115 (c-fms), CD11b and CD11c to identify myeloid / macrophage cells by flow cytometry.

The 3G3 antibody reacts with mouse Foxp3, a 50-55 kDa transcription factor which is a central regulator of T cell activity and is critical for the development and function of regulatory T cells (Tregs). Foxp3 is expressed at constitutively high levels in Treg cells, which are further identified as being CD4+ CD25+. In resting conventional T cells (CD4+ CD25-) Foxp3 expression is restricted, and upon TCR activation is expressed only transiently and in a small proportion of cells. However, the growth factor TGF-beta has been shown to induce expression of Foxp3 in naïve T cells, driving their development into Foxp3+ Tregs, which are called “induced” or “adaptive” Tregs. These cells are phenotypically similar to so-called “natural” Tregs (CD4+ CD25high Foxp3+) which originate in the thymus and comprise the majority of Treg cells. Tregs are critical for maintaining peripheral tolerance and are implicated in the development of autoimmunity. It is important to review the literature in choosing an antibody for the Foxp3 antigen in flow cytometry, as the potential for high background or non-specific staining may be observed. The 3G3 antibody may be used for intracellular detection of Foxp3 in cells from mouse and Rhesus macaque.
