Anti-BrdU Mouse Monoclonal Antibody (FITC (Fluorescein Isothiocyanate)) [clone: PRB-1]
Supplier: Tonbo Biosciences
The fragmentation of genomic DNA by cellular nucleases during the later stages of apoptosis is also one of the most easily measured features of apoptotic cells. Nuclease activity generates DNA fragments ranging from ~300 bp to 50 bp in length, resulting in a typical DNA ‘laddering’ appearance when analyzed by agarose gel electrophoresis. These fragments have exposed 3’-hydroxyl (OH) ends which can be labeled with bromolated deoxyuridine triphosphates (Br-dUTP). An enzyme, terminal deoxynucleotidyl transferase (TdT), is used to catalyze the template-independent addition of Br-dUTP to the 3’-OH ends of double or single stranded DNA. This method is often called TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) or end labeling. Sites where the Br-dUTP is incorporated can then be detected with an antibody specific to BrdU. With the APO-BrdUTM Kit cells are first labeled with Br-dUTP, and then sites of incorporation are detected through staining with a FITC anti-BrdU antibody. Samples can then be analyzed via flow cytometry. Samples that are apoptotic will stain brightly with the anti-BrdU antibody due to the substantial number of exposed 3’-OH sites, while cells that are non-apoptotic will not have incorporated significant amounts of Br-dUTP and will stain dimly. The APO-BrdU Kit is shipped in one container and consists of two packages. Upon arrival one should be stored at 2-8°C and the other at -20°C.

Streptavidin, HRP (Horseradish Peroxidase), Pierce Chemical
Supplier: Thermo Scientific
HRP-Conjugated Streptavidin consists of streptavidin protein that is covalently conjugated to horseradish peroxidase (HRP) enzyme (RZ > 3.0). Streptavidin binds to biotin and the conjugated HRP provides enzyme activity for detection using an appropriate substrate system. This particular product has been used primarily in sandwich ELISA applications to provide consistent measurement of biotinylated detection antibodies.
Formulation: Provided in a conjugate stabilizer containing 0.1% ProClin Compound as a preservative.
Shipped on cold packs.
Application: ELISA: to be used with biotin-labeled detection antibodies. Recommended dilution range is from 1:4,000 to 1:20,000. The optimal dilution must be determined empirically for each specific application. Prepare dilutions immediately before use.

SensoLyte® Red Protease Assay Kit (Fluorimetric), AnaSpec
Supplier: Anaspec Inc
The SensoLyte® Red Protease Assay Kit is widely used for continuous detection of generic protease activities. The kit uses a casein derivative that is heavily labeled with a rhodamine derivative, resulting in almost total quenching of the conjugate's fluorescence. Compared to the SensoLyte® Green Protease Assay Kit, this kit contains a red fluorescent casein derivative. Protease-catalyzed hydrolysis relieves this quenching conjugate, yielding brightly red fluorescent dye-labeled peptides (Ex/Em=546/575 nm). The increase in fluorescence intensity is directly proportional to protease activity. The SensoLyte® protease assay kits do not require any separation steps and can be used to continuously measure the kinetics of a variety of exopeptidases and endopeptidases.

Anti-BrdU Sheep Polyclonal Antibody
Supplier: Genetex
The immunocytochemical detection of bromodeoxyuridine (BrdU) incorporated into DNA is a powerful tool to study the cytokinetics of normal and neoplastic cells. In vitro or in vivo labeling of tumor cells with the thymidine analogue BrdU and the subsequent detection of incorporated BrdU with specific anti-BrdU monoclonal antibodies is an accurate and comprehensive method to quantitate the degree of DNA-synthesis. BrdU is incorporated into the newly synthezised DNA of the S-phase cells and can thus provide an estimate for the fraction of cells in S-phase. Also dynamic proliferative information (such as the S-phase transit rate and the potential doubling time) can be obtained, by means of bivariate BrdU/DNA flow cytometric analysis.

Blocking Solution, Quality Biological
Supplier: QUALITY BIOLOGICAL, INC.
Blocking is a very important step of western blotting, as it prevents antibodies from binding to the membrane nonspecifically. Blocking is often made with 5% BSA or nonfat dried milk to reduce the background. Nonfat dried milk is often preferred as it is inexpensive and widely available. However, milk proteins are not compatible with all detection labels, so care must be taken to choose the appropriate blocking solution.
