DNA Denaturing Solution is useful for denaturing fractionated DNA prior to the transfer from an electrophoresis gel to nitrocellulose or nylon membrane, as well as Northern and Southern blotting.
- Contains: 0.5 M NaOH and 1.5 M NaCl
- Filtered through a 0.22 µm membrane
- Dnase tested
The Denaturing Solution is a strong base, 0.5 M NaOH with 1.5 M NaCl, that denatures double-stranded DNA, so that it can be efficiently transferred to a nylon or nitrocellulose membrane and subsequently hybridized to a labeled probe.
When utilized in conjunction with a depurination solution, treatment with the denaturing solution results in the hydrolysis of the phosphodiester backbone at the sites of depurination. This allows for a more efficient transfer of large DNA in neutral Southern transfers. Following DNA denaturation, the gel must be neutralized by soaking in DNA Neutralization Solution.
