HIC is an excellent technique for the separation of proteins without causing denaturing, therefore scalable from analytical to downstream processing.

Ion exchange separates on the basis of charge on the molecule. It is often used for the separation of charge variants of peptides, proteins and oligonucleotides.
A U/HPLC column with 300Å pore size (or larger) and a C4 or C8 ligand is suitable for small proteins and large peptides. Proteins are retained by adsorbtion of the hydrophobic foot.
