Schoon drinkwater is van essentieel belang voor de gezondheid. In de darmen van zoogdieren en vogels kunnen zowel pathogene als onschadelijke bacteriën worden aangetroffen. Water testen op alle mogelijke typen pathogene bacteriën is een langzaam en duur proces. Daarom wordt water getest op een kenmerkend type darmbacterie - de coliformen - waartoe ook de bekende E. coli behoort. De aanwezigheid van coliformen is een indicatie voor fecale besmetting. In dit experiment testen leerlingen op coliformen in gesimuleerd verontreinigd water met behulp van kleurstoffen en fluorescente reagentia. Dezelfde reagentia kunnen worden gebruikt om watermonsters uit de omgeving te testen. Als extra activiteit kan er een Gram-kleuringstest worden uitgevoerd op de verzamelde monsters.
