Gross cystic disease is a common premenopausal disorder in which gross cysts are the predominant pathologic lesion. It is characterized by production of a fluid secretion which accumulates in the breast cysts. Gross cystic disease fluid is a pathologic secretion from breast composed of several glycoproteins, including a unique 15kDa monomer protein, GCDFP-15. The cells within the body that produce GCDFP-15 appear to be restricted primarily to those with apocrine function. Studies have found GCDFP-15 to be a highly specific and sensitive marker for breast cancer.
GCDFP-15 is expressed by apocrine sweat glands, eccrine glands (variable), minor salivary glands, bronchial glands, metaplasmic epithelium of the breast, benign sweat gland tumors of the skin and the serous cells of the submandibular gland. Breast carcinomas (primary and metastatic lesions) with apocrine features express GCDFP-15. GCDFP-15 is positive in extramammary Paget's Disease. Other tumors tested are negative.

ATP dependent calcium pumps are responsible, in part, for the maintenance of low cytoplasmic free calcium concentrations. The ATP pumps that reside in intracellular organelles are encoded by a family of structurally related enzymes, termed the sarcoplasmic or endoplasmic reticulum calcium (SERCA) ATPases. The SERCA1 gene is exclusively expressed in type II (fast) skeletal muscle. The SERCA2 gene is subject to tissue dependent processing which is responsible for the generation of SERCA2a muscle-specific form expressed in type I (slow) skeletal, cardiac and smooth muscle and the SERCA2b isoform expressed in all cell types. The SERCA3 gene is not as well characterized and is found in non-muscle cells.

Apoptosis is induced by certain cytokines including TNF and Fas ligand (CD95L) of the TNF family through their death domain containing receptors, TNF-R1 and Fas (CD95). Several death receptors including DR3, TRAIL-R1, and TRAIL-R2 were identified. A death domain containing receptor in the TNF-R family was cloned and termed DR6 for death receptor-6. Like TNF-R1, DR6 interacts with death domain containing adapter molecule TRADD. Overexpression of DR6 induces apoptosis and activates NF-κB and JNK. DR6 is widely expressed in human tissues and cell lines. The ligand for DR6 has not been identified yet. T cells from DR6-deficient mice showed preference toward Th2 differentiation in vitro, suggesting that DR6, working through JNK functions to attenuate the Th2 response. Other studies of DR6-targeted mutant mice demonstrated that DR6 functions as a regulatory receptor for mediating CD4(+) T cell activation and maintaining proper immune responses.
