Lipofuscin consists of highly autofluorescent granules of oxidized proteins and lipids that build up in the lysosomes of aging cells. MTT, XTT, and resazurin are reduced by mitochondrial metabolic activity to yield coloured or fluorescent products, and thus are useful for and assaying cell viability and quantitating cell number. MTT and XTT are reduced to coloured formazin salts that can be measured by absorbance. MTT generates an insoluble formazin salt, requiring cell lysis before the absorbance can be measured, while XTT does not require cell lysis for measurement. Resazurin is a non-fluorescent blue dye that is reduced to the pink fluorescent compound resorufin, which can be measured by fluorescence or absorbance.

Recognizes a protein of 30-33 kDa, which is identified as CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood and B cell localization in tissues. It reacts with the majority of B-cells present in peripheral blood and lymphoid tissues and their derived lymphomas. In lymphoid tissue, germinal center blasts and B-immunoblasts are particularly reactive. It is a reliable antibody for ascribing a B-cell phenotype in known lymphoid tissues. Rarely, CD20-positive T-cell lymphomas have been reported. Reactivity has also been noted with Reed-Sternberg cells in cases of Hodgkin's disease, particularly of lymphocyte predominant type.

This antibody is specific to Insulin-like Growth Factor (IGF-1) and shows minimal cross-reaction with IGF-11, Proinsulin, MSF, and Insulin. IGF-1 is a polypeptide growth factor with two isoforms that are produced by alternative splicing. Isoform 1 is also known as IGF-IB while isoform 2 is known as IGF-IA. IGF-1 stimulates the proliferation of a wide range of cell types including muscle, bone and cartilage tissue. It functions as an autocrine regulator of growth. Activation of IGF system has emerged as a key factor for tumor progression and resistance to apoptosis in many cancers like those of breast, thyroid and colon.

Recognizes a protein of 27 kDa, identified as the Bcl-X protein. This MAb shows no cross-reaction with Bcl-2 or Bax protein. Bcl-X has two isoforms, Bcl-XL (long), a 241 amino acid protein which suppresses cell death. And Bcl-XS (short), a 178 amino acid protein lacking a 63 amino acid domain which functions as a dominant inhibitor of Bcl-2. This MAb reacts with both Bcl-XS and Bcl-XL proteins.

MAb IPO-10 defines the antigen, which appears on B cell progenitors following HLA-DR and preceding CD10, CD19, CD22, CD37 and cym. It is expressed on resting B cells and than reappears and persists in cytoplasm and on cell surface until cytoplasmic Ig appears. It is a useful antibody for diagnostics of neoplasms of B cell origins. It reacts with human B cell lines Daudi, Raji, Namalva, EB-3, RPMI-8226 (50% of cells). The MAb does not label T cell lines, blood granulocytes, thymocytes or bone marrow stromal fibroblasts. No significant changes are detected after PHA or ConA stimulation while LPS and PWM stimulated cultures after 18-48h show decreased number of antigen-positive cells but in final terms of cultivation antigen is expressed again. This MAb labels B cell leukemias and some lymphomas. Hairy cell leukemia strongly reacts and 70% of B cell CLL and some B-NHL were also positive. IPO-10 reacts with AMML cells and in a majority of Hodgkin's disease cases a significant percentage of affected lymph node cells were detected.

Recognizes a protein of 80 kDa-90 kDa, identified as CD36. It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. Note that 1-4% of Japanese and East Asia population lack CD36.
