Résistant aux variations de pH, les tampons protègent les échantillons lors de l'ajout d'acides ou de bases. Disponibles sous forme de poudre ou de liquide, ils permettent un suivi efficace des protéines pendant l'électrophorèse et leur séparation. Les tampons salins peuvent également servir à calibrer et à standardiser les pH-mètres pour des mesures précises. Les substances bloquantes empêchent la fixation des anticorps aux sites libres de la membrane, évitant ainsi un bruit de fond parasite.
