Résistant aux variations de pH, les tampons protègent les échantillons lors de l'ajout d'acides ou de bases. Les tampons salins peuvent également servir à calibrer et à standardiser les pH-mètres pour des mesures précises. Les substances bloquantes empêchent la fixation des anticorps aux sites libres de la membrane, évitant ainsi un bruit de fond parasite.
