A protein solubilisation buffer for 2D analysis must solubilise proteins effectively, without disturbing the native charge of the proteins. Urea based solubilisation buffers solubilise proteins effectively, however can modify the native charge of the proteins, due to carbamylation. Urea exists in equilibrium with ammonium cyanate that modifies α- and ε-amino groups, inducing changes in the isoelectric point of proteins leading to artifactual results.

EDTA (ethylenediaminetetraacetic acid) is a polyamino carboxylic acid. It is a colorless, water-soluble solid and is useful as a chelating agent due to its ability to "sequester" metal ions such as Ca2+ and Fe3+.  These cations are responsible for various undesirable effects in molecular biological and cell culture processes. After being bound by EDTA, these metal ions remain in the solution but exhibit diminished reactivity.  The use of EDTA therefore allows for greater control over many experiments.

Sodium Dodecyl Sulfate (SDS) is an anionic detergent with applications in SDS-PAGE protein analysis and nucleic acid purification. It is a wetting agent that is effective in both acid and alkaline solutions, and is most often used as a protein and lipid solubilization reagent. SDS is also a powerful protein denaturant. This 10% solution is prepared in 18 megaohm water and is 0.2µm filtered.
