Highly effective for increasing the intracellular drug accumulation and reversing BCRP-mediated multidrug resistance yet has little activity against P-glycoprotein, multidrug resistance-associated protein (MRP1) and other known drug transporters. In mouse models Ko143 was able to restore the intracellular accumulation and abolish the directionality in net flux of dasatinib. Cells from various organisms increase expression of a class of proteins referred to as heat shock or stress proteins in response to adverse changes in their environment. The members of one class of stress proteins, the Hsp70 family, all bind ATP in vitro, but exist within different intracellular compartments. Members include constitutive form Hsc70 within the cytosol/nucleus; inducible form Hsp70 within the cytosol/nucleus/nucleolus; the constitutive glucose-regulated 78 kDa (or BiP) protein within the lumen of the endoplasmic reticulum; and the constitutive glucose-regulated 75 kDa protein within the mitochondrial matrix. Members of the Hsp70 family appear to function as molecular chaperones, assisting in the folding of other proteins in various intracellular compartments. Grp75 resides in the mitochondrial matrix where it collaborates with Hsp60 in the re-folding of proteins translocated into this organelle. Like itsE. coli homolog DnaK, Grp75 possesses a cation-dependent ATPase activity considered central to its function as a chaperone.

No. BML-GR239), and pro-carcinogenic heterocyclic aromatic amines. Such compounds have been reported to induce Cytochrome P-450 (CYP1A1) in rat liver, human placenta and lung. A rare polymorphism of the CYP1A1 gene has been implicated in familial Parkinson's Disease (PD). In the rat, CYP1A1 has been localized to the globus pallidus, but not to the substantia nigra where cell death associated with the pathology of PD occurs.

Hsp25 is the mouse homologue of the human Hsp27 protein, a member of the small heat shock protein family comprised of a diverse group of proteins from ~15 to >30 kDa. Studies on estrogen-induced proteins in breast cancer cells suggest a direct relationship between elevated stress protein expression and neoplasia. Hsp27 exhibits rapid increased phosphorylation in response to various mitogens, tumor promoters (e.g., phorbol esters) and calcium ionophores1 and elevated levels of Hsp27 correlate with endometrial adenocarcinomas. Interestingly, the regulation of human Hsp27 by estrogens and heat shock resembles the ecdysteroid and heat shock activation of the drosophila Hsp20 genes.
