Die Probe wird im RNA-Solv® Reagenz homogenisiert und lysiert, wodurch die Integrität der RNA erhalten bleibt, während endogene RNasen und andere zelluläre Komponenten aufgeschlossen und denaturiert werden. Die Extraktion des Lysats mit Chloroform denaturiert die Proteine noch mehr und trennt das Gemisch in eine organische und eine wässrige Phase. Die RNA verbleibt ausschließlich in der wässrigen Phase und wird anschließend mit Isopropanol zurückgewonnen.
