Ethidium Homodimer, first developed by Dr. Le Pecq and his colleagues, is a high-affinity fluorescent nucleic acid stain. It binds to both DNA and RNA in a sequence-independent manner and with a >30-fold fluorescence enhancement. The DNA binding of each Ethidium Homodimer covers four base pairs and is believed to occur by intercalation. Because the dye is highly positively charged, it cannot cross cell membranes to stain living cells. It is useful for detecting nucleic acids in solution, or for selectively staining dead cells with damaged plasma membranes.

Histo/Zyme

Histo/Zyme is a stabilized proteolytic enzyme in buffered solution, used to unmask immunoreactive sites altered by fixation and/or the embedding process. Treatment with a proteolytic enzyme in immunohistochemistry (IHC) is required to digest excess linkages and expose the antigen. The use of Histo/Zyme results in superior staining over traditionally used enzyme pretreatments, such as pepsin and trypsin. Moreover, Histo/Zyme requires only a brief incubation at room temperature. Performing antigen recovery with Histo/Zyme will allow you to dilute the primary antibody 5 to 10 fold further and achieve optimal staining.

The incorporation of BrdU into newly synthesized DNA by actively cycling cells is one method for measuring the changing amount of cellular DNA during cell proliferation through each of the cell cycle phases. As a thymidine analog, BrdU is preferentially incorporated into newly replicated DNA which can then be subsequently labeled and analyzed to determine relative DNA content and cell cycle position. Incorporation of BrdU is most commonly detected using anti-BrdU antibodies. A BrdU solution is provided for exposure of actively cycling cells to incorporate BrdU. The EZ-BrdU Kit employs an acid denaturation step. The mild acid method used helps reduce damage to other cellular proteins. After the denaturation step, cells are stained with a FITC anti-BrdU antibody and total DNA is counterstained with a PI/RNase A solution. Two color flow cytometry can then be used to analyze cells that have incorporated BrdU (proliferating cells) in terms of their cell cycle position (G0/1, S, or G2/M phase). The EZ-BrdU Kit is shipped in one container and consists of two packages. Upon arrival one should be stored at 2-8°C and the other at -20°C.

Ethidiumbromid

Ethidium bromide(EtBr) has been used extensively to reduce mitochondrial DNA copy number in proliferating cells and most commonly used in nucleic acid stain for PAGE or agarose gel electrophoresis. The fluorescence of EtBr increases 21-fold upon binding to double-stranded RNA and 25-fold on binding double-stranded DNA so that destaining the background is not necessary with a low stain concentration (10 μg/mL). Ethidium bromide has been used in a number of fluorimetric assays for nucleic acids. It has been shown to bind to single-stranded DNA (although not as strongly) and triple-stranded DNA.4 Because of its ability to bind to DNA, EtBr is an inhibitor of DNA polymerase. A 10mg/mL easy-to-use solution of ethidium bromide in specially filtered, deionized water which is excellent for nucleic acid electrophoresis and purification applications, eliminates the dust hazard associated with powdered ethidium bromide and saves time spent on weighing and mixing. Used in the fluorometric detection of double stranded nucleic acids in gel electrophoresis. Also acts as an RNA polymerase inhibitor, and in separation of high molecular weight DNA's. It confers a deep red stain to DNA by acting as an intercalating agent between the starch bases. Subsequent fluorescence techniques enable the extraction and characterization of DNA bands. Biochem/physiol Actions: Ethidium bromide intercalates double-stranded DNA and RNA and acts as a frameshift mutagen. It can also be used in conjunction with acridine orange to differentiate between viable, apoptotic and necrotic cells.
