Osteopontin (OPN) is an acidic extracellular matrix cell adhesion protein that is relatively abundant not only in bone matrix, plasma, urine, and milk, but is also found in malignant and atherogenic tissues. Phosphorylation, glycosylation and calcium modifications allow intact and fragmented Osteopontin to direct a variety of diverse responses including tissue remodeling, inflammation and cell survival. Plasma Osteopontin has been shown to provide prognostic information in breast, prostate, colon, and lung cancers as well as metastatic carcinomas. The notable presence of Osteopontin in a variety of tumors is strongly correlated to pathological stage, suggesting its critical role in tumor invasiveness, progression and metastasis. In addition, Osteopontin inhibits inducible nitric oxide synthase activity, thereby protecting tumor cells from NO-mediated macrophage cytotoxic attack. Osteopontin is found in atherosclerotic plaques and may drive a number of diabetic vascular pathologies.

The rate limiting enzyme in heme catabolism, heme oxygenase, was first described in 1968 and was thought to be a species of cytochrome P450. However, subsequent work showed the enzyme to be a protein distinct from the P450 family and capable of catalysing the degradation of heme to biliverdin and iron, with the concomitant release of carbon monoxide. This enzyme activity is highly conserved throughout the Animal Kingdom and has been identified in algae and plants as well as in all higher species. Heme oxygenase-1 (HO-1) was first purified to homogeneity from rat liver and pig spleen and shown to have a molecular weight of 32kDa. Increased expression of HO-1 can be induced by a large number of structurally unrelated pharmacological and other agents, including cytokines and oxidants, as well as by heat shock and other forms of cellular stress. Baseline levels of HO-1 expression are high in certain tissues, including spleen and liver, but enriched enzyme preparations are recommended for experimental analysis. Recent data have demonstrated a cytoprotective role for HO-1, preventing cell death by modulating intracellular iron levels.

The proteasome is widely recognised as the central enzyme of non-lysosomal protein degradation. It is responsible for intracellular protein turnover and it is also critically involved in many regulatory processes and, in higher eukaryotes, in antigen processing. The 26S proteasome is the key enzyme of the ubiquitin/ATP-dependent pathway of protein degradation. The catalytic core of this unusually large (2000kDa, 450Å in length) complex is formed by the 20S proteasome, a barrel shaped structure shown by electron microscopy to comprise of four rings each containing seven subunits. Based on sequence similarity, all fourteen 20S proteasomal subunit sequences may be classified into two groups, α and β, each group having distinct structural and functional roles. The α-subunits comprise the outer rings and the β-subunits the inner rings of the 20S proteasome. Observations of the eukaryotic proteasome and analysis of subunit sequences indicate that each ring contains seven different subunits (α7β7β7α7) with a member of each sub-family represented in each particle. Each subunit is located in a unique position within the α- or β-rings. 120S Proteasomes degrade only unfolded proteins in an energy-independent manner, whereas 26S proteasomes degrade native and ubiquitinylated proteins in an ATP-dependent manner. The native protein substrates are recognised by subunits, some with ATP binding sites, of the outer 19S caps of the 26S proteasome. The hybridoma secreting the antibody to subunits HC2, HC3, HC8, HC9, Iota and Zeta was generated by fusion of spenocytes from Balb/c mice which had recieved repeated immunisation with dinitrophenylated proteasomes.
