Immobilised Human Siglec-15, Fc Tag (Cat. No. SG5-H5253) at 10 μg/ml (100 μl/well) on Diamond Protein A Protein, His Tag precoated (0,5 μg/well) plate, can bind Neu5Ac(a2-6)GalNAc-PAA-biotin with a linear range of 0,078 - 1,25 μg/ml (Routinely tested). Immobilised Neu5Ac(a2-6)GalNAc-PAA-biotin on SA Chip can bind Human Siglec-15, Fc Tag (Cat. No. SG5-H5253) with an affinity constant of 4,49 μM as determined in a SPR assay (Biacore T200) (Routinely tested). Loaded Human Siglec-15, Fc Tag (Cat. No. SG5-H5253) on ProteinA Biosensor, can bind Neu5Ac(a2-6)GalNAc-PAA-biotin with an affinity constant of 0,78 μM as determined in BLI assay (ForteBio Octet Red96e) (Routinely tested). Loaded Anti-Siglec-15 MAb (Mouse IgG) on AMC Biosensor, can bind Human Siglec-15, Fc Tag (Cat. No.SG5-H5253) with an affinity constant of 59,7 pM as determined in BLI assay (ForteBio Octet Red96e) (Routinely tested). Human Siglec-15, Fc Tag (Cat. No. SG5-H5253) inhibits Anti-CD3-induced proliferation of PBMC. The ED50 for this effect is 0,46 - 0,54 μg/ml (Routinely tested).
